AnastrozoleAnastrozole

Third-generation nonsteroidal reversible aromatase inhibitor, slightly less potent than letrozole (85 to 90% vs 97 to 99% estradiol reduction). Most commonly used AI in male hormone optimization for estradiol management during TRT.

How it works

Mechanism of action

Reversibly inhibits CYP19A1, blocking aromatization of androgens to estrogens. More titratable than letrozole for managing estradiol in men with lower risk of over-suppression.

Clinical evidence

The research behind Anastrozole.

ATAC Trial: Anastrozole vs Tamoxifen

Lancet (2002)

Improved disease-free survival vs tamoxifen with fewer thromboembolic events and endometrial cancers.

Anastrozole for Hypogonadism in Men

Fertility and Sterility (2001)

1mg/day significantly increased LH, FSH, and testosterone (~50%) while reducing estradiol in young healthy men.
